Current research interests
Since 2009 my work has been mainly focused on the genetic and genomic analysis of bread wheat.
I was involved in the “Physical mapping of wheat chromosome 5A” project from 2009 to 2014, in the frame of the International Wheat Genome Sequence Consortium.
To develop a high quality physical map of 5A, based on two fingerprinted arm-specific BAC libraries, we combined two assembly tools and several anchoring approaches based on PCR marker screening, microarray and BlastN searches, applied to interlinked genomic tools, i.e. genetic maps, deletion bin map, high-density neighbor map, BAC end sequences, genome zipper and chromosome survey sequences. Comparative analysis between the gene distribution along the wheat chromosome 5A and the closest related genomes showed a pattern of syntenic blocks belonging to different chromosomes of Brachypodium, rice and sorghum and regions involving translocations and inversions. The physical map produced thanks to this project is currently the most comprehensive map for 5A chromosome: an essential resource for fine genetic mapping and map-based cloning of agronomically relevant traits, a reference for the 5A sequencing projects, and a fundamental step to gain insight of the evolutionary aspects of bread wheat.
In the frame of the "Canadair" project, a bi-lateral project Italia-Canada, and in collaboration with the group of Hermann Buerstmayr of the Institute for Biotechnology in Plant Production (IFA-Tulln), Austria, I'm working on the genetic and physical analysis of the Fusarium head blight resistance locus on chromosome 5A in bread wheat, employing the genomic resources developed in the “Physical mapping of wheat chromosome 5A” project and moreover exploiting a panel of radiation hybrid lines developed in the reference cultivar Chine spring and specific for 5A chromosome.
I’m also involved in a genome-wide association study for spike traits in an Italian bread wheat collection in collaboration with Research Unit for Cereal Selection in Continental Areas (CRA-SCV), Sant’Angelo Lodigiano (LO, Italy), aimed to dissect these quantitative traits at the genetic levels and to develop molecular markers useful in assisted breeding.
